Most calculators check Mars only from the Ascendant and stop. Classical practice also checks it from the Moon (mind) and Venus (the significator of marriage), and — crucially — applies the cancellation rules (Bhanga). A great many charts that look Manglik are effectively neutralised once those rules are applied.

The body and the life you actually live. Primary, but on its own it over-reports the dosha — this is the only reference most free calculators check.
The mind. Classical practice weighs the Moon above the Lagna, because the dosha is felt as temperament before it is seen as events.
The significator of marriage. A dosha counted from Venus speaks most directly to married life, so it carries the heaviest weight of the three.

Mars then sits in its own sign as lord of the Lagna, so the planet afflicting the house also owns it, and behaves as a benefic for that chart.
Jupiter is the great benefic and the significator of dharma in marriage. Its aspect tempers Mars, which is why this is the most commonly cited cancellation.
